ThePolyATtract®mRNAIsolationSystemsutilizePromega"sMagneSphere®technologytoeliminatetheneedforoligo(dT)celluloseanditsassociatedproblems.WithtotalRNAasthestartingmaterial,thepoly(A)mRNAfractioncanbeisolatedfromothernucleicacidcontaminantsinapproximately45minutes.TheisolatedmRNAissuitableforallmolecularBIOLOGyapplications,includinginvitrotranslationandCDNAsynthesis.
Thesystemusesabiotinylatedoligo(dT)primertohybridizeathighefficiencyinsolutiontothe3´poly(A)regionpresentinmostmatureeukaryoticmRNAspecies.Thehybridsarecapturedandwashedathighstringencyusingstreptavidincoupledtoparamagneticparticlesandamagneticseparationstand.ThemRNAiselutedfromthesolidphasebythesimpleadditionofribonuclease-freedeionizedwater.ThisprocedureyieldsanessentiallypurefractionofmaturemRNAafteronlyasingleroundofmagneticseparation.
