Proposedmodelforb2-AR-andprostanoid-receptor-mediatedPLCandcalciumsignalling.ReceptorscouplingtoGsstimulateAC,resultinginelevatedcAMPlevelsandactivationofEpac1.Epac1thencatalysesGTP-loADIngonRap2B,whichleadstoPLC-eactivation.TheproposedpathwaymayinvolveadditionalsignallingcomponentstoattainPLCstimulation.TheactionofcAMPseemstobeindependentofPKA;Instead,thecAMP-activatedRap-GEFEpacseemstoserveasacAMPeffector,inducingGTPloadingand,hence,activationofRap2B,whichthenleadstospecificactivationofPLC-e,whichhasbeenshowntointeractwithRapGTPases.Itisanattractivehypothesis,therefore,thattheRap-dependentPLCandcalciumsignallingpathwayreportedhereisnotrestrictedtoGs-andAC-coupledreceptors,suchastheb2-ARandtheprostanoidreceptor,butcouldbeusedbyotherreceptorsaswell.
