Thetubbygeneproductisexpressedinthebrainandhasbeenimplicatedbymousegeneticsinobesityandotherdisorderssuchasblindness.Structuralanalysispredictedthatthisgeneactsasatranscriptionfactor,bindingtoDNAtoregulategeneexpression.Morerecently,itwasreportedthattubbybindstophosphoinositidesintheplasmamembrane,helpingtolocalizethisfactorinthemembraneregionintheabsenceofpathwayactivation.TubbyalsointeractswithGalpha-qg-proteinsubunits,localizingtubbytoregionsofg-proteinactivation.ActivationofphosphipaseCactivityandhydrolysisofmembranephosphoinositidesleadstoreleaseoftubbyfromtheplasmamembrane,allowingtubbytoenterthenucleustobindDNAandregulatetranscription.SomeG-proteincoupledreceptorssuchas5-HT2cmaysignalinpartthroughtubbyinadditiontocalcium-mediatedpathwaysandprotein-kinasesignalingpathways.Theidentityofgenesregulatedbytubbyandthemechanismbywhichtubbyaffectsobesityremaintobeexplored.
