ChemokinereceptorsexpressedbyThelpercellshelprecruitcellstospecificlocationsbasedontheirchemoattractantligands.ThepolarizationofTcellsintoTh1andTh2cellsisassociatedwiththeirexpressionofdifferentsubsetsofchemokinereceptors.NaiveCD4positivecellsthathavenotbeenexposedtoantigenexpressCXCR4andCCR7.ThemostabundantchemokinereceptorsonTh1cellsinvolvedinthecellularimmuneresponsetomicrobialagentsincludeCXCR3,CCR1,CCR2andCCR5,whileTh2cellsexpressCCR2,CCR3andCCR5.AnadditionalclassofTcellstermedsemi-na?vecellsmaybeinducedbyTGF-betatoexpressyetanotherclassofchemokinereceptors,CCR4andCCR7.Althoughtheexpressionofchemokinereceptorsindifferentcellsispreferredinthesecases,itisnotabsoluteorexclusive,andthereisoverlapinexpressionbetweenTh1andTh2cells.AlthoughCCR7positivecellshavebeenreportedtolackeffectoraction,notbepolarized,andtohomeonlytolymphoidtissues,otherstudieshavefoundCCR7positiveTh1andTh2cellsandhaveidentifiedCCR7positivecellsinnon-lymphoidtissues.CCR4appearsassociatedwithhomingoflymphocytestotheskin,andCCR9isassociatedwithhomingtothesmallintestine,whileotherchemokineslikeCXCR3andCCR5areassociatedwithhomingtowardinflamedtissuesmorebroadly.TheexpressionofdifferentchemokinereceptorsbysubsetsofThelpercellsplaysanimportantroleinthemigrationandhomingofthesecellsintissues,andtargetingoftheimmuneresponsetospecificcells.
