Micewereinjectedi.p.with0.01mL/gbromodeoxyuridine(AmershamCat.#RPN201).Animalsweresacrificedtwohourslaterandperfusionfixedwith4%paraformaldehyde/PBS.Mammaryglandswereremoved,postfixedin4%paraformaldehydeovernightthenparaffinembeddedandsectionedandmountedontoslides.Alternatively,miceweresacrificedandtheglandsremovedimmediatelywithoutfixationandfrozenforcryostatsectioning.Notsurprisingly,themorphologyisbestontheparaffinsections.(AttachedphotomicrographshowsalabeledductintheaBDominalglandfromapost-pubertalmouse.)II.BrdUDetection
