Description
NefiracetamispyrrolidonederivativethatactsasanagonistatNMDAreceptors,metabotropicglutamatereceptors(mGluR5),andN-typeandL-typevoltage-gatedCa2+channels.Nefiracetamalsopotentiatesacetylcholine(ACh)currentsviaactivityatα4β2nicotinicacetylcholinereceptors(nAChRs).Nefiracetamexhibitsantiepileptic/anticonvulsant,neuroprotective,andcognitionenhancing(nootropic)activities.Invivo,nefiracetamdecreasesseizurefrequencyanddurationandalsoprotectsagainstneuronaldeathinanimalmodelsofpost-ischemicseizure.Inotheranimalmodels,thiscompoundrestoredLTP,improvedspatialmemory,andimprovedobjectrecognitioninseveralbehavioralassays,likelythroughglutamate-dependentmodulationofPKCandCaMKIIsignaling.
