Plasminogen activator inhibitor-1 (PAI-1) Human ELISA Kit
This assay utilises a sandwich Enzyme-Linked Immunosorbent Assay (ELISA) technique and is designed for the quantitative detection of human PAI-1 in plasma or serum in 2.5 hours. A mouse monoclonal antibody specific to PAI-1 has been pre-coated onto a micro-titre plate. The user pipettes standards and samples into the wells and any PAI-1 present is captured by the immobilised antibody. After a short incubation a second biotinylated antibody specific to PAI-1 is added to the wells. After awash step to remove any unbound reagent, streptavidin conjugated to horseradish is added, which binds to the biotinylated detection antibody. Following a wash to remove unbound reagents, an HRP substrate solution is added and colour develops in proportion to the amount of PAI-1 bound initially. The assay is stopped and the optical density of the wells determined using a microplate reader. Since the increases in absorbance are directly proportional to the amount of captured PAI-1, the unknown sample concentration can be interpolated from a reference curve included in each assay.
This Human PAI-1 ELISA kit is designed for quantification of human PAI-1 in serum and plasma. Other biological matrices have not been tested. This kit specifically measures native human PAI-1 and has no cross reactivity to mouse or rat PAI-1.
