Themembranepermeablecompoundisnon-fluorescentuntilitentersviablecells,whereitishydrolyzedbycytoplasmicesteraseenzymestoreleasesthefluorescentamine-reactivedye.Thedyesthencovalentlyreactwithaminegroupsonintracellularproteins,formingfluorescentconjugatesthatareretainedinthecell,whileexcessunreacteddyeiswashedaway. Immediatelyafterstaining,asingle,brightfluorescentpopulationwillbedetectedbyflowcytometry.Witheachcelldivision,daughtercellsinheritroughlyhalfofthefluorescentlabel,allowingthenumberofcelldivisionsthatoccurafterlabelingtobedetectedbytheappearanceofsuccessivelydimmerfluorescentpeaksonaflowcytometryhistogramcomparedtocellsanalyzedimmediatelyafterstaining(Figs.1and2). Cellproliferationdyescanbeusedtotrackcelldivisionsinvivoorinvitro.ThestainingalsocanwithstandfixationandpermeABIlizationforsubsequentimmunostaining.
