Rabbit-on-RodentHRP-Polymerisaone-stepmicro-polymerdetectiondevelopedforusewithrabbitmonoclonal/polyclonalantibodiesonmouseandrattissue.Rabbitprimaryantibodiesonmouseandrattissuesareadvantageousbecauserabbitsecondarydetectionsystemscanbeusedwithminimumcross-reactivitytoendogenousmouseandratIgG.BiocareMedicalhasdevelopedaRabbitHRP-Polymerdetectionsystemwithminimalcross-reactivitytoendogenousmouseandratIgG.Theadvantagesofthispolymertechnologyareincreasedsensitivity,reductionofIHCstepsandtheeliminationofendogenousbiotin.Itcanbeusedwithformalin-fixedparaffin-embeddedtissues,floatingsections,frozensectionsandcellculture.Biocare’snewrabbitpolymerisidealforsinglestainingandcanalsobeusedindoubleandtriplestainingtechniques.Insometissuessuchasspleen,endogenousmouseorratIgGmaybedifficulttoeliminate.BiocareMedicalhasdevelopedanantigenretrievalsolution(RodentDecloaker)thathelpstoreduceoreliminateendogenousIgGandnon-specificbackgroundstaining,andatthesametime,performsantigenretrieval.Temperaturedependentprotocolscanbeusedat80,95and125ºCusingBiocare’sDecloakingChamber.Inaddition,BiocareMedicalhasintroducednewproductstohelpeliminatepersistentratandmouseIgG.TheXMandXRFactoraretwopotentblockersforeliminatingendogenousmouseIgGandratIgG,respectively.Byadding1-2drops(1dropequals33µl)oftheXMorXRFactorto2.5mlofrabbitpolymerdetection,endogenousmouseand/orratIgGwillbereduceddramaticallyorcompletelyeliminated.BiocarehasalsodevelopedRodentBlockMandRodentBlockRthatblockfornon-specificbackgroundstaining,andatthesametime,reduceendogenousmouseandratIgG,respectively.RodentBlockMorRisappliedpriortotheprimaryantibodyfor15to30minutes.
