| Principle | ThisELISAkitusesSandwich-ELISAasthemethod.ThemicroELISAplateprovidedinthiskithasbeenpre-coatedwithanantigenspecifictoHumanAMAIgA.StandardsorsamplesareaddedtotheappropriatemicroELISAplatewellsandcombinedwiththespecificantigen.ThenabiotinylateddetectionantigenspecificforHumanAMAIgAandAvidin-HorserADIshPeroxidase(HRP)conjugateareaddedtoeachmicroplatewellsuccessivelyandincubated.Afterincubation,freecomponentsarewashedaway.Thenthesubstratesolutionisaddedtoeachwell,onlythosewellsthatcontainHumanAMAIgA,biotinylateddetectionantigenandAvidin-HRPconjugatewillappearblueincolor.Theenzyme-substratereactionisterminatedbyaddingStopSolutionandappearsyellowincolor.Theopticaldensity(OD)canbemeasuredspectrophotometricallyatawavelengthof450nm±2nm.TheODvalueisproportionaltotheconcentrationofHumanAMAIgA.TheconcentrationofHumanAMAIgAinsamplescanbecalculatedbycomparingtheODofthesamplestothestandardcurve. |
