| Principle | ThisELISAkitusesSandwich-ELISAasthemethod.ThemicroELISAplateprovidedinthiskithasbeenpre-coatedwithanantibodyspecifictoTSST-1.StandardsorsamplesareaddedtoappropriatemicroELISAplatewellsandcombinedwiththespecificantibody.ThenabiotinylateddetectionantibodiesspecificforTSST-1andAvidin-HorserADIshPeroxidase(HRP)conjugateareaddedtoeachmicroplatewellsuccessivelyandincubated.Afterincubation,freecomponentsarewashedaway.ThentheSubstrateReagentisaddedtoeachwell,onlythosewellsthatcontainTSST-1,biotinylateddetectionantibodyandAvidin-HRPconjugatewillappearblueincolor.Theenzyme-substratereactionwillbeterminatedbyaddingStopSolutionandappearsyellowincolor.Theopticaldensity(OD)canbemeasuredwithspectrophotometryatawavelengthof450nm±2nm.TheODvalueisproportionaltotheconcentrationofTSST-1.TheconcentrationofTSST-1insamplescanbecalculatedbycomparingtheODofthesampleswiththestandardcurve. |
