Asparaginylendopeptidase specifically cleaves peptide bonds on the carboxyl side of asparagine residues, including –Asn-Pro– bonds. The cleavagespecificity of this enzyme has been validated on many peptides and proteins, confirming that bonds locatedon N-terminal or N-second position asparagine residues are not cleaved. Only those bonds adjacent to C-terminal asparagines residues are cleaved. Because asparagine residues bound to sugar chains are not cleaved, this enzyme can be usedfor protein analysisof sequenced glycoproteins. Asparaginylendopeptidase is isolated from Jackbean (Canavalia ensiformis).
