ZR-75-1 Cell Line:
Breast cancer is considered the most prevalent malignancy in females and the fifth most common cause of cancer-related fatalities among women worldwide, representing a significant public health challenge. Metastatic breast cancer is the primary cause of death in patients diagnosed with breast cancer. Preclinical cell line models could help in discovering innovative treatment options for those affected. The ZR-75- cell line is established from a 63-year-old Caucasian female with ductal carcinoma of the breast. This cell line expresses an estrogen receptor and possesses a hypertriploid chromosome number (polyploidy). In addition to producing high levels of MUC-1 mucin mRNA, the ZR-75-1 cell line has a population doubling time of nearly 80 hours. ZR-75 is an epithelial cell line and is useful for studying the properties of tumors in breast cancer patients. Figure 1. Cyclophilin B silencing efficiency was determined by qRT-PCR in the ZR75 cells transfected by Cyclophilin B siRNA or non-silencing siRNA control following the recommended transfection protocol. Cyclophilin mRNA expression levels were measured 48 hours post-transfection. 18S rRNA levels were used to normalize the Cyclophilin B data. Values are normalized to untreated sample. Data are presented as means ± SD (n=6).
Figure 2. Protein expression of Cyclophilin B in ZR-75-1 cells. DNA plasmid expressing Cyclophilin B or siRNA targeting Cyclophilin B were transfected into ZR-75-1 cells following Altogen Biosystems transfection protocol. At 72 hours post-transfection the cells were analyzed by Western Blot for protein expression levels (normalized by total protein, 10 µg of total protein loaded per each well). Untreated cells used as a negative control.
