Alumen450™isaturn-onfluorescentsensor,meaningthatthesolutionfluorescenceincreasesasafunctionoftheconcentrationofdissolvedAluminum.Alumen450™canreADIlybeexcitedoverabroadwavelengthrangefrom280-400nmwithanextinctioncoefficientof8300±600M-1cm-1at318nm.ThefluorescenceemissionisverystronginthepresenceofAluminumions,centeredat ≈420nm.
Alumen450™readychelatesdissolvedAl3+andsubsequentlyshowsanincreaseintheemissionspectraforAlumenBlue™inthepresenceofdissolvedAl3+,i.e.itgetssignificantlybrighter. Theprobeincreasesitsfluorescenceemission>300%ingoingfrom0 →750µMAl3+. Alumen450™showsanalmostlinearresponsetoAl3+from1 →300µMdissolvedAl3+.Alumen450™isalsoahighlyselectiveprobeforboththedetectionanddeterminationofdissolvedAl3+overotherdissolvedcations.
Alumen450™issolddryandissimplyreconstitutedbyaddingwater/MeOH/EtOHand/orbuffer.Werecommenda ≈95/5%ethanol/watermixture(v/v).Onevialofourprobewilltypicallyallowfor10aluminumsensingmeasurementstobeundertaken;50µLofsolventistypicallyaddedtoeachdryvial.Subsequently,ten5µLaliquotsdilutedto3cm3(a3mlvolume)separately,typicallygiveanopticaldensity>0.1at360nm.
Foroptimumperformance,probevialsshouldbestoredat-2to-8°Cwhennotbeingusedand< - 20 °C long term. Probe solutions should be used after dilution.
