BIOLOGicalActivity:RS-1waspreviouslycharacterizedasaRAD51-StimulatorycompoundandnowfoundtobeanHDRenhancertoenhanceCRISPR/Cas9-andTALEN-mediatedknock-inefficiency.Itincreasestheknock-inefficiencybytwo-tofive-foldatdifferentloci,whereasNHEJinhibitorSCR7hasminimaleffects.RS-1wasappliedforanimalproductionandhaveachievedmultifoldimprovementontheknock-inratesaswell.RS-1canstimulatethehumanhomologousrecombinationproteinRAD51invariousconditions.InPC3prostatecancercells,RS-1–inducedlethalitywasaccompaniedbytheformationofmicroscopicallyvisIBLeRAD51nuclearproteinfocioccurringintheabsenceofanyDNA-damagingtreatment.TreatmentwithRS-1promotedsignificantantitumorresponsesinamousemodel.HowtoUse:Invitro:RS-1wasusedat15µMfinalconcentrationinTALEN-orCas9-mediatedknock-inexperiments.Invivo:RS-1wasusedat7.5 μMfortheinvivoknock-inanimalproduction.RS-1canbedosedtoanimalbyIPinjectionat110mg/Kgonceperdaytogetantitumorresponses.Reference:
