Thekithasbeendevelopedtoovercomethecommondifficultiesuniquetoneuronalsamplesincludingthefragilenatureofbraintissuesections,highbackgroundproblems,poorcounterstainingwithcommondyes,andtheneedtoperformduallabelingexperimentstodetectcellspecificantigensinconjunctionwithapoptoticcells.AkeyfeatureisNeuroPore™,aproprietarypermeABIlizationreagentthatgentlypermeabilizessampleswhileretainingcellmorphology.NeuroPore™alsocontainsblockingreagentstoallowitsuseasanantibodydiluentinimmunohistochemistryandtoreducebackgroundstaining.
DNAfragmentsgeneratedbyapoptosisareend-labeledwithmodifiednucleotidesusingahighlypurifiedterminaldeoxynucleotidyltransferaseenzyme(TdT).TheincorporatednucleotidesaredetectedusingahorserADIshperoxidasesystemthatcatalyzestheconversionofdiaminobenzidine(DAB)intoavisIBLedarkbrownprecipitate.NeuroTACS™IIcontainstheBlueCounterstaintoallowvisualizationofallcellswithinthesamplewithgoodcontrasttothebrownDABprecipitate.ADABenhancerisalsoprovidedwiththekitfortheoptiontointensifyordarkenDABstaining.Inaddition,theBlueCounterstainiscompatiblewiththered-coloredsubstratesusedforphosphatasedetectionindoublelabelingexperiments.Theprotocolincludesdetailsforlabelinginsituforapoptosisandantigendetectiononthesamesample.Toensurethatyourownsampleshavebeenprocessedandpermeabilizedcorrectly,weprovideTACS-Nuclease™,auniquereagentusedtogenerateapositivecontrolwithyourownsamples.Thisprovidesahighdegreeofconfidencefordatainterpretationandcanhelppinpointproblemstepsinthelabelingprocedure.
