IntheSuperoxideDismutaseAssay,ionsgeneratedfromtheconversionofxanthinetouricacid,andhydrogenperoxidebyxanthineoxidase(XOD),convertNBTtoNBT-diformazan.NBT-diformazanabsorbslightat550nm.SODsreducesuperoxideionconcentrationsandtherebylowertherateofNBT-diformazanformation.TheextentofreductionintheappearanceofNBT-diformazanisameasureofSODactivitypresentinexperimentalsamples.Theassayisfreeofinterferencebyothercatalyticactivities,andisidealforassayingSODinmammaliancelllysates.Thekitcontainstheproperlysisbufferandthereagentsneededfor100experimentaltests,50positivecontrols,and50negativecontrols.UnlikesomeotherassaykitsforSOD,thissystemisnotgreatlydisturbedbytracemetals.Eachassayrequiresonlyaboutfiveminutes,andafterasimplecalculation,thepercentinhibitionoftheformationofNBT-diformazanbySODisconvertedtotherelativeactivityofthesample.
