A water soluble carbodiimide also called EDAC or EDCI, commonly obtained as the hydrochloride. Typically, it is utilized in the pH range 4.0-6.0 without buffers. In particular, amine and carboxylate buffers should be avoided. It can be used as a chemical crosslinker for collagen, reacting with the carboxylic acid groups of the collagen polymer which can then bond to the amino group in the reaction mixture.1,2Efficiency of EDC-mediated coupling is increased in the presence of Sulfo-NHS.3Excess reagent and crosslinking byproducts are easily removed by washing with water or dilute acid. Crosslinking can be done in physiologic solutions without adding organic solvent. Applications for EDC include conjugate carboxyl to amine groups in peptides and proteins;4,5convert carboxyls to amine-reactive Sulfo-NHS esters;6,7crosslink proteins to carboxyl coated beads or surfaces;8Activate nanoparticles with amine-reactive Sulfo-NHS esters;9couple haptens to carrier proteins (e.g. attach a peptide to KLH);10DNA labeling through 5’ phosphate groups.11
