Introduction: Cas9(CRISPRassociatedprotein9)isaRNA-guidedDNAendonucleaseenzymeassociatedwiththeCRISPR(ClusteredRegularlyInterspacedShortPalindromicRepeats)adaptiveimmunitysystemin Streptococcuspyogenes.TheCRISPR/Cas9systemisthelatestRNA-guided,endonucleasetoolingenomeeditingwhichallowsforveryspecificgenomicdisruptionandreplacement.Guidedbyatarget-specific,singleguideRNA(sgRNA),theCas9nucleaseproteinservestounwindthegenomicDNAduplexandcleavebothstrandsuponrecognitionofthetargetsequencebythesgRNA,resultingindouble-strandedbreaks.Theresultingdouble-strandedbreakgetsrepairedbythenon-homologousendjoining(NHEJ)pathway,leADIngtoadisruptionintheopenreadingframeofthetargetedgene.
