ProductDescription

CytoForm®-200consistsofa4.5x4.5x0.15cm3DprintedscaffoldmadeoutofHyperelasticBone™(trademarkpropertyofDimensionInx®).ItcomesasalargesheetthatcanbereADIlycutorpunchedouttomeetexperimentalneeds.

TheHyperelasticBone™scaffoldsconsistof~90%Hydroxyapatiteintheformof1-30µmsphericalparticles,and~10%polylactide-co-glycolide(PLG).

Thesescaffoldsprovideinterconnectedporosity(forvascularization),highbioactivity,easyhandling,workABIlityandhighmechanicalstability.Thesefeaturesmakethesubstratesidealforcellcultureinboneandhardtissueregeneration.

TheCytoForm®-200scaffoldsareethanolwashedpriortoshipment,butarenotconsideredsterile.

EveryotherlayerisoffsetintheXandYby0.5mmtoprovideoptimalcell-seedingsurfacearea.

CytoForm®-200ischemicallystableinneutralpH,isflexIBLe,butalsohascompressive/mechanicalstrength.

Sterilization

Itisrecommendedtocutorpunchsamplesintendedfortheexperimentpriortosterilization(ie.4mmbiopsypunch).

  1. Washsamplesin70%ethanolfor30-60minutes.
  2. Rinsesamples3-5timesinsterilewaterorPBS.
  3. Removeexcessliquidfromscaffoldsviadabbingonfilterpaperorsterilizecloth,oraspiratingoutexcessliquid.Donotallowscaffoldstobecomecompletelydrypriortocellseeding.

CellSeedingInstructions

  1. Placewettedscaffoldsintodesiredtissuecultureplate.
  2. Dispense10uLofcellsuspensionontothescaffoldfora4-5mmdiameterscaffold(~50,000cells).
  3. Allowcellstoattachfor30-60minutespriortoaddingadditionalmedia.
  4. Slowlyfillthecellculturewellwithmediaandincubateat37°C

CellSeedingInstructionsUsingCollagenHydrogel

  1. Placewettedscaffoldsintodesiredtissuecultureplate.
  2. Addcellstoaneutralizedcollagensolution(suchasPureCol®EZGel).
  3. Slowlydispensethecollagen/cellsolutionoverthescaffoldandallowthesolutiontofullypermeatethroughoutthescaffold.
  4. Allowcollagentopolymerizewithinthescaffoldfor60-90minutesat37°C.
  5. Fillthewellwithcellculturemediumuntilthescaffoldiscompletelycoveredandcontinueincubatingat37°C.]
