Qdot®655streptavidinconjugatecomprisesabiotin-bindingprotein(streptavidin)covalentlyattachedtoafluorescentlabel(Qdot®nanocrystal).Streptavidinhasaveryhighbindingaffinityforbiotin,andaconjugateofstreptavidiniscommonlyusedtogetherwithaconjugateofbiotinforspecificdetectionofavarietyofproteins,proteinmotifs,nucleicacids,andothermolecules(forexample,abiotinylatedprimaryantibodyboundtoaproteintargetcanbedetectedwithafluorescentlylabeledstreptavidin).Strategiessimilartothisareusedinmanydetectionprotocolsincludingwesternblots,flowcytometry,imagingandmicroscopy,andmicroplateassays,andarealsousedinpurificationworkflowstoachievetargetedfractionation.Qdot®nanocrystalconjugatesareprovidedas1µMsolutions.

PropertiesofQdot®Nanocrystals
TheQdot®streptavidinconjugateisthesizeofalargemacromoleculeorprotein(~15–20nm)andrepresentsthebrightestclassofstreptavidindetectionreagents.Qdot®streptavidinconjugatesaremadefromananometer-scalecrystalofasemiconductormaterial(CdSe),whichiscoatedwithanadditionalsemiconductorshell(ZnS)toimprovetheopticalpropertiesofthematerial.TheQdot®705andQdot®800streptavidinconjugates,whichincludeCdSeTe,aremadeinasimilarfashion.Thiscore-shellmaterialisfurthercoatedwithapolymershellthatallowsthematerialstobeconjugatedtoBIOLOGicalmoleculesandtoretaintheiropticalproperties.
