ThePierceQuantitativeColorimetricPeptideAssayprovidesmodifiedBCAreagentsforthereductionofCu+2toCu+1andaproprietarychelatorthathavebeenoptimizedforthequantitationofpeptidemixtures.Inthisreaction,thecopperisfirstreducedbytheamidebackboneofpeptidesunderalkalineconditions(Biuretreaction).Theproprietarychelatorthencoupleswiththereducedcoppertoformabrightredcomplexwithabsorbanceof480nm.Thesignalproducedfromthisreactionis3-4foldmoresensitivethantheMicro-BCAProteinAssayforpeptideanalysis.
Thekitalsocontainsahighqualitypeptidedigestreferencestandardforuseingeneratinglinearstandardcurvesandtoserveasacalibrationcontrol.Thiscolorimetricpeptideassayrequiresasmallamountofsample(20µL)andhasaworkingpeptideconcentrationrangeof25–1000μg/mL.Theincreasedsensitivity,lowsampleassayvolume,andincludedreferencestandardareessentialforaccurateandrobustmeasurementofpeptidedigestsamples,especiallyformassspectrometryapplications.
