AntibodiestotheSSAantigen(alsoknownasRoantigen)areoneofthemostfrequentSEROlogicalMarkersofautoimmunityinrheumaticdiseases.Theyappearin60-70%ofpatientswithSjögren’ssyndrome(SS),30-40%ofpatientswithsystemiclupuserythematosus(SLE),and3-5%ofpatientswithrheumatoidarthritis(RA).TheSSAantigeniscomprisedofanacidic60kDaproteinthatmayexistincomplexwithRNA(80-112bases).However,RNAisnotrequiredforSSAantigenicity.TheSSAantigenispredominantlycytoplasmic,andnotlocatedinthenucleus.Therefore,patientswithantibodytoSSAmaybeANAnegativeonroutinetesting.Approximately62%ofANAnegativelupuspatientshaveantibodiestoSSA.Twotypesofanti-Ro/SSAantibodieshavebeenidentified;Ro/SSAantigensof60kDaand52kDa.Anti-SSA-60kDaantibodiesarelinkedtocertaindisorderssuchasSS,SLE,neonatallupusandcongenitalheartblock.Clinically,thepresenceofaSSA52hasbeenreportedinawidevarietyofdiseases,includesinflammatorymyositis,primarybiliarycirrhosisandSS.Principle:
AutoimmuneELISAkitsmeasureautoimmuneantibodiesintheserum.Itisbasedontheprincipleofasolidphaseenzyme-linkedimmunosorbentassay.Theassayutilizesaspecificantigenforimmobilizationonthemicrotiterwellsandanti-humanIgGantibodyconjugatedtohorserADIshperoxidase(HRP)fordetection.Thetestsampleisallowedtoreactsimultaneouslywiththetwocomponents,resultingin autoimmuneantibodiesbeingsandwichedbetweenthesolidphaseandenzyme-linkedantibodies.Afterincubation,thewellsarewashedtoremoveunbound-labeledantibodies.AHRPsubstrate,TMB,isaddedtoresultinthedevelopmentofabluecolor.ThecolordevelopmentisthenstoppedwiththeadditionofStopSolutionchangingthecolortoyellow.Theconcentrationofautoimmuneantibodies isdirectlyproportionaltothecolorintensityofthetestsample.Absorbanceismeasuredspectrophotometricallyat450nm.
