UnlikeconventionalreversetranscriptionwhichneedstotalRNA,SignosishasdevelopedaCL(TM)cDNAsynthesis-basedplatearray.ItcansynthesizecDNAprobesthroughreversetranscriptiondirectlyincelllysateswithoutRNApreparation.ThesynthesizedcDNAprobesarethenappliedforaplatearrayhybridization.Targetedgenesarespecificallycapturedontoindividualwellsonaplatethroughapre-coatedgene-specificoligonucleotide.ThecapturedcDNAsarefurtherdetectedwithstreptavidin-HRP.Luminescenceisreportedasrelativelightunits(RLUs)onamicroplateluminometer.Theexpressionlevelofgenesisdirectlyproportionaltotheluminescentintensity.
