Background
Autophagy,theprocessofbulkdegradationofcellularproteinsthroughanautophagosomic-lysosomalpathwayisimportantfornormalgrowthcontrolandmaybedefectiveintumorcells.Itisinvolvedinthepreservationofcellularnutrientsunderstarvationconditionsaswellasthenormalturnoverofcytosoliccomponents.Beclin-1,aprincipalregulatorofautophagosomeformation,isinturnregulatedbyAmbra1.Ambra1associateswithBeclin-1througharegionnearitscenterasdeterminedbyyeasttwo-hybridassay.Nullmutationsinthisgeneinmiceresultedinembryoniclethalitywithsevereneuraltubedefectsassociatedwithautophagyimpairment,accumulationofubiquitinatedproteins,unbalancedcellproliferationandexcessiveapoptoticdeath.FurThermore,down-regulationofAmbra1inculturedcellsthoughRNAinterferencedecreasedthelevelofrapamycin-andnutrientstarvation-inducedautophagy.MultipleisoformsofAmbra1areknowntoexist.
