Alpha-tubulinbelongstothetubulinsuperfamily,whichiscomposedofsixdistinctfamilies.Alongwithbeta-tubulins,alpha-tubulinsarethemajorcomponentsofmicrotubules.Thesemicrotubulesareinvolvedinawidevarietyofcellularactivitiesrangingfrommitosisandtransporteventstocellmovementandthemaintenanceofcellshape.Alpha-andbeta-tubulindimersareassembledto13protofilamentsthatformamicrotubuleof22-nmdiameter.TyrosineligaseaddsaC-terminaltyrosinetomonomericalpha-tubulin.AssembledmicrotubulescanagainbedetyrosinatedbyaCytoskeleton-associatedcarboxypeptidase.Anotherpost-translationalmodificationofdetyrosinatedalpha-tubulinisC-terminalpolyglutamylation,whichischaracteristicofmicrotubulesinneuronalcellsandthemitoticspindle.LikeGAPDHandbeta-Actin,thisantibodymakesanexcellentloADIngcontrolinimmunoblots.
