Background
Aldh3A2isamemberofthealdehydedehydrogenasesuperfamily,agroupofNAD(P)(+)-dependentenzymesthatcatalyzetheoxidationofawidespectrumofaliphaticandaromaticaldehydes.Aldehydedehydrogenaseenzymesarethoughttoplayamajorroleinthedetoxificationofaldehydesgeneratedbyalcoholmetabolismandlipidperoxidation.Aldh3A2catalyzestheoxidationoflong-chainaliphaticaldehydestofattyacids.MutationsintheAldh3A2genecauseSjogren-Larrsonsyndrome,aninheritedneurocutaneousdisorder.Patientswiththisdisorderdisplayichthyosis,mentalretardationandspasticdiplegia.Thepathogenesisofthecutaneousandneurologicalsymptomsisthoughttoresultfromabnormallipidaccumulationinthemembranesofskinandbrain,theformationofaldehydeSchiffbaseadductswithamine-containinglipidsorproteins,ordefectiveeicosanoidmetabolism.
