Background
Theadhesionanddegranulationadaptorprotein(ADAP)wasinitiallyidentifiedasamolecularadapterthatcouplesTcellreceptor(TCR)stimulationtotheAvidityofintegrinsgoverningTcelladhesion.TCRstimulationpromotestheformationofamulti-proteincomplexcontainingCARMA1,MALT1,andBCL-10,whichthroughtheassociationofADAP,ultimatelyactivatestheNF-kBfamilyoftranscriptionfactors.MorerecentexperimentshaveshownthatADAPcontrolsoptimalTcellproliferation,cytokineproduction,andexpressionoftheBcl-2familymemberBcl-x(L),suggestingthatADAPregulatesTcellactivationbypromotingantigen-dependentTcell-antigenpresentingcell(APC)activation.AtleastthreeisoformsofADAPareknowntoexist.
