Thiscanbealleviatedbyfirstimmunoprecipitatingtheantibody:antigencomplexandthendetectingtheantigen.Thismethodresultsinaverycleanandstrongsignal. BothAng-1andAng-2proteinshavepredictedmolecularweightsofapproximately57kDaandappearonwesternblotsclosetotheirpredictedmolecularweights. Insomeinstancesadditionalbandsmaybeseenatapproximately75kDawhichrepresenthighlyglycosylatedformsoftheproteinthatmigrateatahigherapparentmolecularweight.
Purity/Specifity
Antiopoietin1isdirectedagainstmouseangiopoietin1andshowsnoreactivitywithmouseangiopoietin2. A57kDabandisdetectedwhenassayedbyimmunoblot. Thisreagentcross-reactswithhumanangiopoietin1. Partiallycrossreactivityisnotedwithhumanangiopoietin2.
