ThepolyclonalantibodyreactswithratC5L2.TheorphanreceptorC5L2isaseventransmembranereceptorwith40%aminoacidsequenceidentitytotheC5areceptor,CD88.Expressionhasbeenfoundongranulocytesandimmaturedendriticcells.IncontrasttoCD88,C5L2isuncoupledfromGproteinsduetoananaminoacidreplacementofargininebyleucineinthesocalledDRYregionattheendofthethirdintracellulartransmembranedomain.FollowingC5abinding,C5L2seemsneithertoinduceclassicalsignallingnortocauseBIOLOGicalresponses.WhenC5L2wastransfectedintoseveralcelltypes,C5afailedtoinducechemotaxis,degranulation,orinteracellularcalciummobilization.ThehighaffinfityofC5L2forC5aanditsmetabolite,C5adesArgandalowaffinityforC3aandC3adesArgsuggestthatthisreceptorisascavengerofcomplementfragmentsinvolvedinthedefenceagainsttheharmfulsideeffectsofcomplementactivation.C5L2expressiononhumanneutrophilsisknowntobedecreasedinsepsis;lossofexpressionmaybeaprognosticindicatorinthisconditionassurvivorsofsepsistendtohavehigherC5L2levels.
