Thepolyclonalantibodyrecognizeshumanindoleamine2,3-dioxygenase(IDO).IDOisanintracellularheme-containingenzymethatcatalyzestheoxidativecleavageoftheindoleringofseveralimportantregulatorymolecules,liketryptophan,SEROtoninandmelatonin.Bydoingthis,IDOinitiatestheproductionofBIOLOGicallyactivemetabolites,commonlyreferredtoaskynurenines.IDOiswidelyexpressedinavarietyofhumantissuesaswellasinmacrophagesanddendriticcells(DCs).Ininflammation,interferons(IFNs)actonspecificreceptorstotriggerIDOinduction.TheproductionofIFN-gammaandinductionofIDOrepresentimportantantimicrobialmechanisms.DegradationanddepletionoftryptophanbyIDOinhibitsthegrowthofviruses,bacteriaandparasites.FurThermore,IDOplaysacomplexandcrucialroleinimmunoregulationduringinfection,pregnancy,autoimmunity,transplantation,andneoplasia.Bylocaldepletionoftryptophanandincreasingproapoptotickynurenines,IDOgreatlyaffectsT-cellproliferationandsurvival,bothinvitroandinvivo,andalsoaffectsB-cellandNK-cellfunctionandsurvival.ThereisacentralroleforIDOexpressionintoleranceinvolvingregulatorycellsandDCs.IDOactsasanintermediatepathwayinLPS-inducedproductionofreactiveoxygenspeciesandNF-kappaBactivation,twoprocessesthatleadtoDCmaturation.ThepolyclonalantibodyisobtainedafterimmunizationofsheepwithrecombinantIDO.ReactivityofthepolyclonalantibodywithIDOhasbeenconfirmedonimmunoblotwithIFN-gammastimulatedhumanperipheralbloodlymphocytes.
