TheOX-2,orCD200receptor(CD200R),belongstotheimmunoglobulinsuperfamily.CD200RcontainstwoIg-likeextracellulardomains,oneIg-likeC2-type(immunoglobulin-like)domainandoneIg-likeV-type(immunoglobulin-like)domain.andmediatesinhibitorysignalsinmyeloidcells.MicedeficientfortheligandCD200developenhancedexperimentalallergicencephalomyelitisandcollagen-inducedarthritis.UnlikeCD200R,theCD200R-likereceptorsfunctionasactivatingreceptors.TheycontainshortcytoplasmictailsandalysineresidueinthetransmembraneregionandarelikelytosignalviaadaptorproteinssuchasDAP12,DAP10,FcRγ,orCD3ζ.
CD200R3isexpressedinuterusandbonemarrow-derivedmastcells(atproteinlevel).Itisexpressedinuterus,spleen,bonemarrow-deriveddendritic,basophilandmastcells.CD200R3isalsoexpressedinthelungofN.brasiliensis-infectedmice.Itisweaklyexpressedinbrain,testis,lungandthymus.
CD200R3isexpressedpreferentiallyoncellsofthemyeloidlineage,includingmastcellsandbasophils.Bothcelltypesareinvolvedinthehostdefensesystemagainstpathogensandinthedevelopmentofallergicdisorders.Theyfunctionasindependentessentialinitiatorsofallergicreactions.Basophilsaretheleastcommonleukocytesintheperipheralbloodaccountingapproximately0.5%ofallleukocytes.InvivodepletionofbasophilsbyusingmonoclonalantibodyBa103provedtheessentialroleofbasophilsinthedevelopmentandmaintenanceofIgE-mediatedchronicallergicinflammation.
TheactivatingCD200receptors,likeCD200R3,playanimportantroleinIgE-independentmastcellandbasophilactivation.MonoclonalantibodyBa103activatesbasophilsexvivoandelicitssystemicanaphylaxiswhenadmiNISTeredinvivo.Cross-linkingofCD200R3onMC/9cellsbymonoclonalantibodyBa103inducesdegranulation.FurThermore,monoclonalantibodyBa103isusefulfordepletionofbasophils.Thisantibodydoesnotcross-reactwithCD200R,CD200R2,CD200R4orCD200R5.
