1.IntendedUse TheGenWayVarizellazosterIgGAntibodyELISATestKithasbeendesignedforthethedetection andthequantitativedeterminationofspecificIgGantibodiesagainstVarizellazosterinserumand plasma.FurtherapplicationsinotherbodyfluidsarepossIBLeandcanberequestedfromthe TechnicalServiceofGenWay. Thisassayisintendedforresearchuseonly. 

2.GeneralInformation VarizellaisanacuteviraldiseasecausedbytheVarizellazostervirus(VZV)thatbelongstothe herpesvirusfamily. Thediseasebeginswithfeverandmalaiseandamaculopapularrashthatchangeswithinhoursto vesiclesthatremainduring3to4days.Lesionspustulate,crustandhealslowlyovera3weekperiod.Lesionsappearinthescalp,face,neck,trunk,mucousmembranesofthemouthandupperrespiratorytract.Sometimes,especiallyinadults,thefeverandgeneralclinicalstatuscanbesevere, butthediseaseisrarelyfatal.Themostcommoncauseofdeathinadultsistheprimaryviralpneumonia,andinchildren,secondarybacterialinfectionsorcentralnervoussysteminvolvement.Childrenwithacuteleukemiaorimmunocompromisedpersonsareathigherriskofageneraldisseminationofthediseasewithfataloutcome.Newbornsthatareinfectedafter5to10days,andtheones bornfrommothersthatwereinfected5daysbeforeortwodaysafterbirth,candevelopseveregeneralisedVarizellawithamortalityrateof30%.Infectionatthebeginningofpregnancyrarelygives congenitalmalformations. ReactivationofVZVisusuallymanifestedaszoster.Zosternormallybeginswithunilateralsharp andwelllocalisedpain.InthiscasethetiteroftheIgGantibodiesisincreased,whereasthetiterof IgMantibodiesisraisedonlyinsomefewcases. 

3.PrincipleoftheTest TheGenWayVarizellazosterIgGantibodytestkitisbasedontheprincipleoftheenzyme immunoassay(EIA).Varizellazosterantigen isboundonthesurfaceof themicrotiterstrips. Dilutedsampleserumorready-to-usestandardsarePipettedintothewellsofthemicrotiterplate.A bindingbetweentheIgGantibodiesoftheserumandtheimmobilizedVarizellazosterantigentakes place.Afteraonehourincubationatroomtemperature,theplateisrinsedwithdilutedwash solution,inordertoremoveunboundmaterial.Thenready-to-useanti-human-IgGperoxidase conjugateisaddedandincubatedfor30minutes.Afterafurtherwashingstep,thesubstrate(TMB) solutionispipettedandincubatedfor20minutes,inducingthedevelopmentofabluedyeinthe wells.Thecolordevelopmentisterminatedbytheadditionofastopsolution,whichchangesthe colorfrombluetoyellow.Theresultingdyeismeasuredspectrophotometricallyatthewavelength of450nm.TheconcentrationoftheIgGantibodiesisdirectlyproportionaltotheintensityofthe color.
