IL-8(alsoknownasNAP-1forNeutrophil-activatingpeptide)isachemoattractantproteinforneutrophils.Thiscytokinebelongstoanewfamilyofchemotacticpeptidescalled"chemokines".Thisproin¬ammatorymediatorissecretedbydierentcellssuchasmonocytes,neutrophils,endothelialcells,broblastafteractivation,andbymitogen-stimulatedTlymphocytes.TheroleofIL-8intherecruitmentofneutrophilsinthelungduringARDShasalsobeensuggested.
Principlesofthemethod:TheGenWayIL-8-EASIAisasolidphaseEnzymeAmplifiedSensitivityImmunoassayperformedonmicrotiterplate.Theassayusesmonoclonalantibodies(MAbs)directedagainstdistinctepitopesofIL-8.Calibratorsandsamplesreactwiththecapturemonoclonalantibody(MAb1)coatedonmicrotiterwellandwithamonoclonalantibody(MAb2)labelledwithhorserADIshperoxidase(HRP).Afteranincubationperiodallowingtheformationofasandwich:coatedMAb1-humanIL-8-MAb2-HRP,themicrotiterplateiswashedtoremoveunboundenzymelabelledantibody.Boundenzymelabelledantibodyismeasuredthroughachromogenicreaction.Chromogenicsolution(TMB)isaddedandincubated.ThereactionisstoppedwiththeadditionofStopSolutionandthemicrotiterplateisthenreadattheappropriatewavelength.Theamountofsubstrateturnoverisdeterminedcolourimetricallybymeasuringtheabsorbance,whichisproportionaltotheIL-8concentration.AcalibrationcurveisplottedandIL-8concentrationinsamplesisdeterminedbyinterpolationfromthecalibrationcurve.TheuseoftheEASIAreader(linearityupto3ODunits)andasophisticateddatareductionmethod(polychromaticdatareduction)resultinahighsensitivityinthelowrangeandinanextendedcalibrationrange.
StorageandStABIlity:Beforeopeningorreconstitution,allkitscomponentsarestableuntiltheexpirydate,indicatedontheviallabel,ifkeptat2to8C.
Unusedstripsmustbestored,at2-8C,inasealedbagcontainingadesiccantuntilexpirationdate.
