Background:
HumanGFAP-δ,alsotermedGFAP-ε,isanalternativesplicevariantoftheGFAPprotein.GFAPproteinsareselectivelylocatedinastrocytesandrepresentthemajorconstituentofastrocyticintermediatefilaments.GFAPexpressionlevelsarehighlyvariableduringdevelopmentofthecentralnervoussystem.GFAP-δdiffersfromthepredominantspliceformofGFAP,GFAP-α,initsC-terminalsequenceinwhichtheoriginalaminoacidsequence,encodedbyexons8and9,arereplacedbyauniqueandconservedtailof42aminoacidsencodedbyexon7+andcalledtheGFAP-δtaildomain.ThisGFAP-δtaildomainisabletobindpresenilinproteinsofwhichmutatedformsareacauseofAlzheimerdisease.GFAP-δdoeshowevernotdiscriminatebetweenAlzheimerandcontrolfissue.UpregulationofGFAP-δhasanegativeeffectonGFAPfilamentformation.IncontrasttoGFAP-α,GFAP-δisnotupregulatedinastrogliosis.Source:
