Specificity:
Theenzymehasbroadspecificity,cleavingα(1-2,3,6)-linkedmannose,althoughsomekineticpreferencehasbeenobserved(α1-2,6>3).Theenzymewillnotcleaveasingleα(1-6)linkedmannoseresiduefromcoreβ-mannose,butwill,however,removeasingleα1-3linkedmannosefromthecoreβ-mannose. Byusingenzymeconcentrationsofaround50U/mlandextendedincubationtimes(upto18hours)at37°C,completeremovalofallnon-reducingterminalαlinkedmannoseresiduesmaybeachieved.
Toexpediteglycansequencingstudies,thesluggishactivityofGKX-5010JackBeanαmannosidasetowardα(1-6)-linkedmannoseresiduescanbeovercomebyusingtheenzymeincombinationwiththeαmannosidasefromXanthomonasmannihotis whichrapidlycleavesα(1-6)linkages.TheX.mannihotisα-mannosidase mayinhibittheactionofothermannosidasesifabranched(α1-6)mannoseispresentinthesubstrate,soistypicallyaddedafterincubationofthesubstratewithJackbeanα-mannosidase(GKX-5010).
