Specificity:
Theenzymereleasesα(2,3)-,α(2,6)-,α(2,8)-,andα(2,9)-linkedN-acetylneuraminicacidfromcomplexcarbohydrates.Theinitialrateofhydrolysisofα(2,6)linkagesisreportedtobeapproximatelytwicethatofα(2,3)-linkedsialicacidhowever,inpractice,thiskineticselectivityisoflittleconsequenceduringextendedincubations.Effectivedigestionofglycolipidsubstratesisfacilitatedbyadditionofadetergent,suchassodiumtaurodeoxycholatetotheincubation.
SialidaseAiscapableofreleasingN-glycolylneuraminicacid(Neu5Gc,NGNA)inadditiontoN-acetylneuraminicacid(Neu5Ac,NANA)[1],althoughsimilarlytoothersialidases[2]theactivityislowertowardNeu5GcthanNeu5Ac.
Theenzymecanalsobeusedtoremovesialicacidfromgangliosides[3],glycosphingolipds(ceramide&oligosaccharide)withsialicacid.
MaybeusedforexoglycosidasesequencinginconjunctionwithSialidaseS(GK80021),whichisspecificforα(2,3)-linkedsialicacids.
