Interleukin-12(IL-12,IL12)is,whenBIOLOGicallyactive,aheterodimer(p70)consistingoftwocovalentlylinkedsubunits,p35andp40.IL-12isproducedbyantigen-presentingcells,specificallydendriticcellsandmacrophages.IL-12isaproinflammatorycytokinethatpromotesTh1-typeresponsesbyinducingIFN-γproductionandenhancingproliferationandcytotoxicityofNKandTcells.Interleukin-23(IL-23)isadisulfide-linkedheterodimerwhichconsistsofauniquep19subunitandthep40subunitofIL-12.IL-23isaproinflammatorycytokine,whichisproducedbymacrophagesanddendriticcellsuponinfectionswithcertainpathogens.TheIL-23receptorisexpressedonTh17cellsandIL-23isresponsIBLeforTh17expansionandstABIlization.SincebothIL-12andIL-23heterodimerscomprisethep40subunit,captureimmunoassaysbasedonantibodiestop40willdetectbothcytokines.Suchassayscanalsodetectp40monomersanddimers.
