Genisteinisanisoflavoneandaninhibitoroftyrosineproteinkinases. IthasbeenreportedtoinducetopoIIdependentcleavages(Biochem.Pharmacol.39:737-740[1990]). TopoGENprovidesthisreagentasacontrolpoison/inhibitoroftopoisomeraseII(Note,GenisteinmayalsoinhibittopoisomeraseI.) Forinvitrocleavagedetection,werecom-mendthatthedrugbetestedoverawiderangeofuMconcentrations;however,cleavagesarestronglyinfluencedbytheamountoftopoisomeraseinthereactionaswellasthetypeofassaybeingemployed(SDSK+vs.RADIoactiveEndLabeledDNAvs.testingforplasmidcleavages). Notethatlargeamountsofenzymewillberequiredtodetectcleavagecomplex,comparedtoassaysthatmeasurecatalyticactivity.
