Likeβ-lactams,cephalosporinsinterferewithPBP(penicillinbindingprotein)activityinvolvedinthefinalphaseofpeptidoglycansynthesis.PBP’sareenzymeswhichcatalyzeapentaglycinecrosslinkbetweenalanineandlysineresiduesprovidingadditionalstrengthtothecellwall.Withoutapentaglycinecrosslink,theintegrityofthecellwallisseverelycompromisedandultimatelyleadstocelllysisanddeath.Resistancetocephalosporinsiscommonlyduetocellscontainingplasmidencodedβ-lactamases.Likemanycephalosporins,ceftiofurisresistanttoβ-lactamases.
