BACKGROUND
Ethidiumbromide(EB)foundfameinthelate1940sasanantitrypanosomal,antimicrobial,antibacterial,andantiviralagent.ItsBIOLOGicaleffectsareadirectconsequenceoftheinhibitionofnucleicacidsynthesis,whichinturnisrelatedtothespecificbindingofthedrugtoDNA.EBinhibitsDNApolymeraseandbindsinvitrotobothRNAandDNA.InvestigationintotheprecisenatureoftheDNA-EBbindingmechanismledtothediscoverythatEBbindsbyamechanismtermedintercalation.Thisprocesshasbeenstudiedextensivelyduringthepastthreedecades,andthephotophysicalchangesthataccompanyintercalationhavebeensuccessfullyappliedtoquantitateandstructurallyelucidateDNA.MorerelevanttotheuseofEBinmolecularbiologyaretheobservedvariationsinitsfluorescentcharacteristicsonbindingtopolynucleotides.
