Tightjunctions,orzonaoccludens,formacontinuousbarriertofluidsacrosstheepitheliumandendothelium.TheyfunctioninregulationofparacellularpermeABIlityandinthemaintenanceofcellpolarity,blockingthemovementoftransmembraneproteinsbetweentheapicalandthebasolateralcellsurfaces.ZonaoccludensproteinsZO-1,-2,and-3(alsoknownasTJP1,2,and3)areperipheralmembraneadaptorproteinsthatlinkjunctionaltransmembraneproteinssuchasoccludinandclaudintotheactinCytoskeleton.ZO-1and-2arerequiredfortightjunctionformationandfunction.Insubconfluentproliferatingcells,ZO-1andZO-2havebeenshowntocolocalizetothenucleusandplayaroleintranscriptionalregulation,possIBLythroughfacilitatingnuclearimport/exportoftranscriptionalregulators.TheZO-2geneistranscribedfromtwopromoters,generatingtheZO-2AandZO-2Cisoforms.ZO-2Clacksa23aminoacidamino-terminalsequencefoundinotherZO-2isoforms.Whilebothisoformsappeartobewidelyexpressed,abnormalregulationoftheZO-2genemaybecorrelatedwithdevelopmentofductalcancer.
