Thelymphaticvasculatureformsasecondcirculatorysystemthatdrainsextracellularfluidfromthetissuesandprovidesanexclusiveenvironmentinwhichimmunecellscanencounterandrespondtoforeignantigen.RecentlyanumberofinterestingmoleculeshavebeenidentifiedthatmaybeexploitedasMarkersforlymphaticendothelium,includingthehyaluronanreceptorLYVE1,PALE,VEGFR3,andpodoplanin.LYVE1hasbeenidentifiedasamajorreceptorforHA(extracellularmatrixglycosaminoglycanhyaluronan)onthelymphvesselwall.ThededucedaminoacidsequenceofLYVE1predictsa322-residuetypeIintegralmembranepolypeptide41%similartotheCD44HAreceptorwitha212-residueextracellulardomaincontainingasingleLinkmodule,theprototypicHAbindingdomainoftheLinkproteinsuperfamily.LikeCD44,theLYVE1moleculebindsbothsolubleandimmobilizedHA.However,unlikeCD44,theLYVE1moleculecolocalizeswithHAontheluminalfaceofthelymphvesselwallandiscompletelyabsentfrombloodvessels.Hence,LYVE1isthefirstlymph-specificHAreceptortobecharacterizedandisauniquelypowerfulmarkerforlymphvesselsthemselves.
