IgDcross-linkingonBcellsisanidealmethodfortheassessmentofacutepolyclonalBcellactivationacrossmultiplestrainsofmice.Followinganti-IgDinjection(100μl/mouse,i.v.),peripheralBcellsarerapidlyactivatedasevidencedbyup-regulationofCD86andCD69.ActivatedBcellswillsecreteeffectorcytokines(IL-4)andacquireeffectorfunction(secretionofIgEantibodies)within5-7days.Anti-mouseIgDantibodycanalsobeusedtoinducefunctionalBcellsinvitro.ThismodelsystemisidealforspecificexaminationofkeypathwaysandplayersinthefunctionalactivationofperipheralBcellsinvivo.
