TheImmunoStarN-terminalneuronalnitricoxidesynthaseantiserumwasqualitycontroltestedusingstandardimmunohistochemicalmethods.Theantiserumdemonstratesstronglypositivelabelingofrathypothalamus,striatum,cortexandspinalcordusingindirectimmunofluorescentandbiotin/avidin-HRPtechniques.Recommendedprimarydilutionsare1/1,000-1/2,000inPBS/0.3%TritonX-100-biotin/avidin-HRP.
ByWesternblotanalysisofbrainhomogenatestheantibodyspecificallylabelsabandofapproximately155kD.Immunolabelingiscompletelyabolishedbypre-adsorptionwithsynthetichumannNOS(134-148)at5µgpermLofdilutedantibody.NocrossreactivitywithotherformsofNOSwasobserved.
