Theantibodywascharacterizedbyimmunohistochemistry.ImmunohistochemicalstainingofratbraincorrelateswellwithNorthernblotanalysis,insituhybridizationandreceptorautorADIographystudies.Immunolabelingiscompletelyabolishedbypreadsorptionwithsyntheticrat5-HT7receptor(8-23).BlastPdatabasesequencehomologysearchesindicatethattheaminoacidsequenceisuniquetorat5-HT7A,5-HT7Band5-HT7C.Thereisalsosignificantsequenceoverlapwiththemouseandhumanforms.
PhotoDescription:IHCimageofneuronsstainingforthe5-HT7receptorintheratbraincortex.Thetissuewasfixedwith4%formaldehyde/0.05%glutaraldehydein0.1Mphosphatebuffer,beforebeingremovedandpreparedforvibratomesectioning.FloatingsectionswereincubatedatRTin10%goatseruminPBS,beforestandardIHCprocedure.Primaryantibodywasincubatedat1:500for48hours,goatanti-rabbitsecondarywassubsequentlyaddedfor1hourafterwashingwithPBS.Lightmicroscopystainingwasachievedwithstandardbiotin-streptavidin/HRPprocedureandDABchromogen.
