TheImmunoStar5-HT2Creceptorantiserumwasqualitycontroltestedusingstandardimmunohistochemicalmethods.Theantiserumdemonstratesstronglypositivelabelingofratchoroidplexusandhippocampususingindirectimmunofluorescentandbiotin/avidin-HRPtechniques.Recommendedprimarydilutionsare/500-1/1000inPBS-Bn/Av-HRPtechnique.Intensificationmethodssuchasnickelwillapproximatelydoublethedilutionfactorasrecommended.
TheantibodywascharacterizedbyimmunohistochemistryandWesternblot.Westernblottingrevealedasinglebandofapproximately70kD.
Preincubationoftheantibodywithanexcessofthesyntheticpeptideblockedstaining.ImmunohistochemicalstainingofratbraincorrelateswellwithNorthernanalysis,insituhybridizationandreceptorautorADIography.BlastPdatabasesequencehomologysearchesconfirmedthatthissequenceisuniquetorat,mouseandhuman5-HT2Creceptors.
