Theantiserumdemonstratesstronglypositivelabelingofratcortex,arcuateandhippocampususingindirectimmunofluorescentandbiotin/avidin-HRPtechniques.Recommendedprimarydilutionsare1/200-1/300inPBS-Bn/Av-HRPdetection.Intensificationmethodssuchasnickelwillapproximatelydoublethedilutionfactorasrecommended.
TheantibodywascharacterizedbyimmunohistochemistryandWesternblot.Westernblotshowedasinglebandofapproximately45kDusingadilutionof1/100.Thewesterninformationisincludedasspecificityinformation.Preincubationoftheantibodywithanexcessofthesyntheticpeptideblockedstaining.ImmunohistochemicalstainingofratbraincorrelateswellwithNorthernanalysis,insituhybridizationandreceptorautorADIography.
