Description:

GlutathioneS-transferase(GST)isafamilyofenzymesthatplayanimportantroleindetoxificationofxenobiotics.GSTcatalyzestheformationofthethiolgroupofglutathionetoelectrophilicxenobiotics.Itutilizesglutathionetoscavengepotentiallytoxiccompoundsincludingthoseproducedasaresultofoxidativestressandispartofthedefensemechanismagainstthemutagenic,carcinogenicandtoxiceffectsofsuchcompounds.TheGSTFluorometricActivityAssayKitprovidesasimple,fluorescence-basedinvitroassayfordetectingtheGSTactivityusingfluorescenceplatereader.Theassayutilizesmonochlorobimane(MCB),adyethatreactswithglutathione.ThefreeformofMCBisalmostnonfluorescent,whereasthedyefluorescesblue(ex/em=380/461nm)whenreactswithglutathione.GSTcatalyzestheMCB-glutathionereactionsandthefluorescencelevelsareproportionallytotheamountsofGSTpresenceinthereaction.Thus,theGSTlevelinsamplescanbeeasilydetectedusingafluorometerora96-wellfluorometricplatereader.ThekitcandetectGSTactivityincrudecelllysateorpurifiedproteinfraction,andalsoquantitateGST-taggedfusionprotein.Detects<0.5mU.
