TrkBistheprimaryreceptorforBDNF(brain-derivedneurotrophicfactor.TrkBalsobindsNT4andNT3butlessefficiently.Uponligand-binding,thereceptorundergoeshomodimerization,autophosphorylationandactivation.TrkBactivationrecruits,phosphorylatesand/oractivatesseveraldownstreameffectorsincludingSHC1,FRS2,SH2B1,SH2B2andPLCG1thateachregulatedistinctoverlappingsignalingcascadeswithincells.ThroughSHC1,FRS2,SH2B1,SH2B2,theseactivatetheGRB2-Ras-MAPKcascadethatregulates,forinstance,neuronaldifferentiationincludingneuriteoutgrowth.ThesesameeffectorsalsocontroltheRas-PI3kinase-AKT1signalingcascadethatmainlyregulatesgrowthandsurvival.TrkB,viaactivationofPLCG1andthedownstreamproteinkinaseC-regulatedpathways,alsocontrolssynapticplasticity,andthusplaysaroleinlearningandmemorybyregulatingbothshorttermsynapticfunctionandlong-termpotentiation.PLCG1alsoleadstoNF-Kappa-Bactivationandthetranscriptionofgenesinvolvedincellsurvival.OnesuchconsequenceisthatPLCG1activationviaTrkBisabletosuppressanoikis,theapoptosisresultingfromlossofcell-matrixinteractions.(Reference:www.uniprot.org) |
